No, jokers can only be used in combinations of 3 or more identical tiles (pung, kong, etc). Jokers cannot be used on singles or pairs.
Keep in mind that combinations like NEWS or years (eg: 2022) are considered 4 single tiles, so jokers cannot be used in them.
